1 ILLUMINA (NextSeq 550) run: 5.8M spots, 1.8G bases, 730.1Mb downloads

Design: Genomic libraries were prepared using DNA extracts and the Nextera XT DNA Library Preparation kit (Illumina, San Diego, CA, USA) according to the manufacturers protocol. Libraries were quality checked using an Agilent 2100 Bioanalyzer and DNA High Sensitivity kit and then pooled in an equimolar ratio. The pool was gel purified using a 2% agarose gel and the Qiagen QIAquick gel extraction kit (Qiagen, Germantown, MD, USA). Following purification, the pool was sequenced on an Illumina NextSeq 550 instrument using a Mid-Output v2.5 chemistry 300-cycle kit to produce 2x150 bp reads.

The genomes examined in this study were sequenced within the framework of an extended Course-based Undergraduate Research Experience (eCURE). To enhance our understanding, we employed several enrichments involving potassium iodate, potassium iodide, manganese carbonate, and manganese sulfate. Following enrichment, we identified specific isolates through partial sequencing of the 16S rDNA gene and characterized their ecophysiology. Subsequently, these selected isolates underwent whole-genome sequencing and assembly, carried out by undergraduate students involved in the research.
